I want to make a new flower/shrub bed in an area of lawn.
What is the best way to get rid of the grass and prevent the grass and weeds growing back up through the bed?.

To convert an area of lawn to make a new bed, there are a number of options.

You can dig the sods and turn down the green end thus burying it at spade-depth and killing the grass.

Or you can cover the grass with old carpet or black polythene for a few months to kill the grass and then plant.

Or you can apply Roundup, or similar glyphosate-based weedkiller, to kill the grass and plant directly into the dead sod. Normally some bark mulch is used to cover the dead grass, which can be unsightly until it rots away.
